Drug Design, Synthesis and Biological Evaluation of New Neuroprotective Agents Potentially Useful in Brain Diseases

Among  motoneuron diseases Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disorder of devastating impact that causes injury and death of lower motor neurons in the brainstem and spinal cord, and of upper motor neurons in the motor cortex. A series of novel substituted 2-aminobenzothiazoles and 4H-3,1-benzothiazine and 2H-1,4-benzothiazine derivatives which proved to reduce glutamate release in in vitro assays was recently patented. Supported by molecular modeling simulations, the synthesis of differently functionalized series of benzothiazoles and benzothiazines, with the aim to obtain new selective inhibitors of neuronal nitric oxide synthase (n-NOS) is in progress.
